I would like to know if there is a possibility in the page builders grid system to use more specific column classes for different screen dimensions. Bootstrap realizes this with their different col-lg-*, col-md-* etc. classes which can be combined together.
If there is no solution in page builder the only way to realize this is only to setup media query rules manually, right?

Unfortunately, this isn’t currently possible without a fair amount of custom coding. I’ve marked this thread as a feature request. We often look through these threads to get ideas for what to add in our future updates. Hopefully, we’ll be able to add this in a future update.
I’m not too sure what your specific use case is but based on your question, it’s quite likely you will need to use media queries. To make this easier, you could apply classes to rows or cells and apply CSS based on if they’re present or not. You can apply a class to a row by opening the row (click the wrench icon above the row) and head over to the row styles sidebar. Open the Attributes settings group and set the Row Class as desired.
