I am in need of advice. I would like to upload a 272MB promotional video to our homepage. Is there a preferred widget for optimal playback? What are my next steps here?

With this size, I’d suggest using a 3rd party service like Vimeo or YouTube. They’ll handle all the complicated things like CDN, encoding, optimizing, etc. Generally, hosting your own videos only really works for smaller videos.

If you host your video using one of these services, then you can just paste the video URL either in your main editor or in a Visual Editor widget inside Page Builder. WordPress will handle the rest – like fetching the embed code.
